Abstract
Gold nanoclusters (AuNCs) are an emerging class of gold nanostructures with unique properties, such as high photostability and large Stokes shift. This confers them high potential as alternatives to organic dyes and quantum dots in bioimaging. Nevertheless, their low colloidal stability and luminescence quantum yield have limited their full potential. To overcome such limitations, encapsulation of the AuNCs in several host materials was proposed. In this thesis, we studied the encapsulation of AuNCs in polymers and silica nanoparticles using several methodologies. The thesis is divided in several sections: the first one is a comprehensive bibliographic revision of the encapsulation of AuNCs in different host materials. The second deals with the encapsulation of the AuNCs of 25 Au atoms stabilized by mercapto hexanoic acid (MHA-AuNCs) in methacrylate polymer nanoparticles of 50 nm diameter by photo-miniemulsion polymerization. The third section address the encapsulation of AuNCs by polymerization induced self-assembly (RAFTPISA). The hairy core-shell nanoparticles with a poly(butyl acrylate) (PBA) core and a poly (Nisopropylacrylamide) (PNAM) shell encapsulating in situ MHA-AuNCs and AuNCs stabilized with hexanethiol (MH-AuNCs). The hybrid hairy nanoparticles, with a hydrodynamic diameter of 50 nm, maintain their colloidal and optical properties for at least 1 year. They were imaged in HeLa cells, showing their internalization into the cell cytoplasm. The fourth section is the direct synthesis of AuNCs using thiol terminated PNAM homopolymers and copolymers. By changing the molecular weight of the PNAM-SH, NIR emitting AuNCs with hydrodynamic diameter between 5 and 16 nm were synthesized. The in cellulo tests in HeLa cells showed their incorporation into the cytoplasm. The synthesis of polystyrene (PS) stabilized AuNCs was also explored, which were later encapsulated into PS-co-PEO micelles by solvent exchange. The NIRemitting PS-AuNCs were encapsulated into the micelles by bulk and microfluidics nanoprecipitation, leading to particles with 215 nm and 125 nm, respectively. The fifth section studies the one-pot encapsulation of AuNCs with NIR emission in mesoporous silica nanoparticles of about 50 nm. In a second approach, the AuNCs were conjugated with MnFe2O4 NPs in a core-shell system, with a MnFe2O4 NP in the core and a mesoporous silica shell incorporating the AuNCs. The core-corona nanostructures combine in a single particle the superparamagnetic behaviour (MS = 13.4 emu g-1 at 300 K) of the core with the NIR AuNCs emission from the corona. Finally, a last section summarizes the conclusions and points the challenges and opportunities of the work and points to future developments namely concerning the application of the nanostructures in bioimaging.
