Abstract
There is a commercial interest in finding enzymes to be included in detergents as biodegradable ingredients that increase their efficiency. In this work, a library of bacteria from moderate and extreme aquatic environments was screened for lipases using the rhodamine B method, a fluorescent indicator that allows the detection of free fatty acids. Selected strains were cultivated at 30°C in saline media (19.4 g/L NaCl) and the activity of the cells was determined using tributyrin and trioctanoin with a high-throughput method. This consisted of following the reaction in a spectrophotometer with a pH indicator, phenol red, and measuring the absorbance decrease that occurs with the release of fatty acids. The maximum activity observed for tributyrin was 1.988 nmol free fatty acids/(min.mg biomass) for strain 705 grown in MB, while for trioctanoin it was 0.966 nmol free fatty acids/(min.mg biomass) for strain 729 grown in TSB. The strain 729 was identified as a Pseudoalteromonas undina. The maximum growth rate for P. undina decreased from 0.847 h-1 to 0.296 h-1 when the cells grew on olive oil as carbon source. However, there was a 22% increase in activity for tributyrin, in contrast to the effect on activity for trioctanoin, where it decreased by 64%. In the presence of detergent, the activity increased 30% and 80% when tributyrin and trioctanoin were used as substrates, respectively. Finally, the kinetics of the enzyme followed the Substrate Inhibition Model.
